摘要: 【背景】 海草床是重要的“蓝碳”生态系统,对全球碳汇有重要贡献。海草床沉积物剖面的垂直梯度特征显著,表层呈现氧化态,富含活性有机质,而深层呈还原态,以惰性有机质为主。【目的】 探究这种垂直特征如何影响微生物的丰度和群落分布。【方法】 利用荧光定量PCR和16S rRNA基因高通量测序等手段,测定了山东荣成天鹅湖大叶藻海草床不同深度(5、10、15、20、25和30 cm)沉积物中细菌和古菌丰度、多样性和群落结构的变化。【结果】 细菌和古菌16S rRNA基因拷贝数随深度的增加而降低,在沉积物5 cm深处,细菌的16S rRNA基因拷贝数显著高于20 cm和30 cm层(ANOVA,P < 0.05)。深度对细菌和古菌α多样性指数没有显著影响(P > 0.05)。细菌中相对丰度最高的是变形菌门,其次是绿弯菌门,拟杆菌门,浮霉菌门等,其中δ-变形菌和浮霉菌的相对丰度随深度显著增加(P < 0.05)。古菌群落中深古菌门比例最高,在25 cm深处达到70%以上;其次是乌斯菌门、洛基古菌门、广古菌门和奇古菌门等。奇古菌门比例随深度增加而显著降低(P < 0.05),其他古菌类群在不同深度间差异不显著(P > 0.05)。【结论】 海草床沉积物细菌和古菌的丰度、多样性和群落分布具有明显的垂直特征,这种特征可能受沉积物有机质组成和氧化还原状况影响。

Abstract: [Background] The seagrass meadow was a "Blue carbon" ecosystem contributing greatly to global carbon sequestration in coastal sediments. The vertical profile of seagrass sediments exhibits a strong redox gradient, where the surface layer was oxic and rich in labile organic matters and the deeper layer was reduced and dominated by recalcitrant organic matters. [Objective] We hypothesized that bacterial and archaeal communities varied greatly in abundance and community structure along the vertical gradient in the seagrass sediments. [Methods] Quantitative real-time PCR and high-throughput sequencing were applied to characterize the prokaryotic communities at different sediment depths (5, 10, 15, 20, 25 and 30 cm) in the Zostera marina dominated meadow. [Results] The 16S rRNA gene copy numbers of bacteria and archaea decreased with the increasing sediment depth, and the bacterial copy numbers in the 5-cm layer was significantly higher than those in the 20-cm and 30-cm layers (ANOVA test, P < 0.05). Depth had no significant effect on bacterial and archaeal α diversity indices (P > 0.05). The most dominant bacterial phylum was Proteobacteria, followed by Chloroflexi, Bacteroidetes, and Planctomycetes. The relative abundances of δ-Proteobacteria and Planctomycetes were significantly increased with increasing depth (P < 0.05). Bathyarchaeota was the most dominant archaeal phylum, accounting for 70% in the 25-cm layer. Other abundant archaeal phyla were Woesearchaeota, Lokiarchaeota, Euryarchaeota and Thaumarchaeota. The relative abundance of Thaumarchaeota decreased significantly with increasing depth (P < 0.05). [Conclusion] The benthic archaeal and bacterial communities in the seagrass meadow sediments exhibited obvious vertical characteristic, which could be driven by organic matter composition and sediment redox status.

海草床(seagrass meadows)是生产力和生物多样性最高的典型近海生态系统之一[1-2],其与红树林和盐沼并称为“蓝碳”系统[3],对近海有机碳埋藏和增汇具有重要意义[4-5]。有研究指出,海草床增加了未来海洋CO2的短期极端值和长期抵消量[6],对缓解海洋酸化有重要意义。此外,海草床还具有其他重要的生态服务价值,如改善水体透明度、控制浅水水质、抗波浪潮,并为许多种类的海洋生物提供生存场所和食物等[7]

海草床沉积物是营养盐再生与转化发生的重要场所。大量海草的叶和根茎组织残片积累在沉积物中,其分解再生的营养物质对维持海草生产力具有重要作用[8]。然而,由于海草组织中纤维素含量很高(超过50%),不易降解,大部分会随沉降作用埋藏到沉积物深处[9]。海草具有发达的根状茎,其中的气腔组织能够将地上叶组织光合作用产生的氧气输送到根际,并最终渗透到根际沉积物中[10-11]。另外,海草能够截留水体中的有机颗粒如浮游微藻等,加之新鲜根组织还会向沉积物中释放光合作用产生的小分子活性有机碳[12],因此,海草床沉积物剖面呈现出表层氧化态且富含活性有机质,以及深层还原态以惰性有机质为主的垂直梯度特征。这种生物地球化学梯度势必影响微生物种群和生态功能的垂直分布。

我们之前的研究工作发现,海草覆盖区表层沉积物中δ-变形菌(δ-Proteobacteria)和绿弯菌(Chloroflexi)相对丰度较高,而裸露区沉积物中则是拟杆菌(Bacteroidetes)较高,推测沉积物有机质的组成是影响海草床沉积物细菌组成的主要因素[13]。在红树林沉积物中,Wu等[14]发现绿弯菌(Chloroflexi)的相对丰度随沉积物深度的增加而增加,而拟杆菌(Bacteroidetes)随沉积物深度的增加而降低;Pan等发现[15],深古菌(Bathyarchaeota)在红树林沉积物20-38 cm层相对丰度最高,其剖面分布与沉积物总有机碳含量和pH显著相关[15-16]。然而目前海草床沉积物剖面的微生物分布特征尚不清楚。

本研究在山东荣成天鹅湖自然保护区内的大叶藻(Zostera marina)海草床采集沉积物柱状样品,通过荧光定量PCR和16S rRNA基因高通量测序和分析技术,研究沉积物垂直剖面细菌和古菌的拷贝数、多样性和群落结构,并分析了其垂直变化的规律,研究结果可为进一步了解海草床微生物介导的营养转化过程及保护和修复海草床提供参考。

1 材料与方法 1.1 样品采集

研究区域位于山东省威海市荣成县天鹅湖自然保护区(122o34′26.96′′E,37o21′1.46′′N)的大叶藻海草床,分别于2015年12月(column 1、column 2)和2016年3月(column 3、column 4)利用PVC柱状采样管(直径5 cm)各采集2根约35 cm深的沉积物柱样。现场严格按每5 cm切割分层,并取每一层上部约1 cm厚的沉积物样品作为测试样品,最终每根沉积物柱分别获得6层(即5、10、15、20、25与30 cm)样品(表层至深层依次标号为D5、D10、D15、D20、D25和D30),共成功获得23个沉积物分层样品(缺失2015年12月column 2的D30样品),即每一层3-4个平行样。沉积物样品混匀后置于5 mL的冻存管中,立即保存于液氮中,回到实验室之后转移到-80 ℃冰箱保存,并于1个月之内完成环境因子的测定和DNA的提取。

1.2 主要试剂和仪器

土壤DNA提取试剂盒,MO Bio Laboratories公司;荧光定量PCR试剂盒,TaKaRa公司。激光粒度仪,Mastersizer公司;元素分析仪,Elementar公司;电感耦合等离子体质谱仪,PerkinElmer公司;连续流动分析仪,SEAL分析有限公司;微量核酸测定仪和荧光定量PCR仪,Thermo Fisher Scientific公司;HiSeq测序平台,Illumina公司。

1.3 沉积物理化因子分析

沉积物粒径(grain size)采用激光粒度仪测定,总有机碳(total organic carbon,TOC)和总氮(total nitrogen,TN)利用元素分析仪测定,重金属(Pb、V、Cr、Mn、Fe、Co、Ni、Cu、Zn、As和Cd)的含量由电感耦合等离子体质谱仪测定。铵氮(NH4+),硝态氮(NO3-)和亚硝态氮(NO2-)浓度利用2 mol/L KCl超声(400 W,工作30 s,间歇30 s,共1 h)萃取后,于2 000 r/min离心15 min收集上清液,再利用连续流动分析仪测定。

1.4 DNA的提取和高通量测序

沉积物DNA的提取按照试剂盒说明书进行,并利用微量核酸测定仪测定其含量。分别利用古菌引物对U519F (5′-CAGYMGCCRCGGKAAHACC-3′)/ 806R-Arch (5′-GGACTACNSGGGTMTCTAAT-3′)[17]和细菌引物对341F (5′-CCTAYGGGRBGCASCA G-3′)/806R-Bact (5′-GGACTACNNGGGTATCTAA T-3′)[18]通过PCR扩增古菌16S rRNA基因的V4区和细菌16S rRNA基因的V3–V4区[17-18],然后利用Illumina HiSeq高通量测序平台进行双端测序(诺禾致源生物信息科技有限公司)。

1.5 荧光定量PCR

细菌和古菌的绝对丰度采用SYBR Green荧光定量PCR法获得,在ABI 7500 Fast平台上进行。PCR反应体系(20 μL):SYBR Green I Mix 12.5 μL,正、反向引物(20 μmol/L)各0.5 μL,样品模板DNA 2 μL,ddH2O 4.5 μL。细菌和古菌荧光定量PCR的扩增引物对分别为341F (5′-CCTACGGGAGGCAG CAG-3′)/519R (5′-GWATTACCGCGGCKGCTG-3′)[19]和931F (5′-AGGAATTGGCGGGGGAGCA-3′)/M110R (5′-BGGGTCTCGCTCGTTRCC-3′)[20]。PCR反应条件:95 ℃ 7 min;95 ℃ 30 s,64 ℃ (古菌)或56 ℃ (细菌) 30 s,72 ℃ 30 s,共40个循环;最后按照ABI 7500 Fast实时荧光定量检测系统默认程序制备熔解曲线:95 ℃ 15 s,60 ℃ 1 min,95 ℃ 30 s,60 ℃ 15 s,检测是否存在非特异性扩增。每个样品设置3个重复,并利用不含DNA的去离子水作为阴性对照。利用含有细菌和古菌16S rRNA基因全长的质粒构建标准曲线,稀释梯度为原浓度的10-2-10-8。所有PCR反应的扩增效率均在90%以上,相关系数(R2)均大于98%。

1.6 高通量数据分析

高通量数据分析基于Linux平台的QIIME 1.9.0流程[21],参考数据库为SILVA (version 128)[22]。具体流程如下:首先进行双端测序结果拼接,并利用Mothur (version 1.39)软件[23]去除前后引物错配碱基 > 1、同聚体 > 6、序列长度 < 200的序列;然后利用VSEARCH程序[24],参考SILVA数据库去除嵌合体,并选取97%的序列相似度作为阈值划分OTU;利用BLAST法(E-value=10-5)对细菌和古菌进行系统分类,然后去除Singleton和非古菌/细菌序列;最后对获得的OTU丰度表,利用软件R包“edgeR”进行标准化。

1.7 数据处理

应用SPSS 20统计软件中的单因素方差分析(analysis of variance,ANOVA)检验不同深度沉积物样品间细菌和古菌16S rRNA基因拷贝数、α多样性及主要类群(相对丰度 > 1%)相对丰度的差异,并利用最小显著差法(least-significant difference,LSD)进行多重比较不同层间差异;基于OTU的距离矩阵利用PRIMER 6.0软件对细菌和古菌进行非度量多维尺度(non-metric multidimensional scaling,nMDS)排序,并利用相似性分析(analysis of similarities,ANOSIM)检验沉积物深度对细菌和古菌群落结构的影响。

2 结果与分析 2.1 沉积物理化性质的垂直特征

由于2015年12月的沉积物样品不足,只对2016年3月12个样品进行了理化因子测定,结果见表 1。从整体上看,沉积物总氮(TN)含量对深度不敏感,范围在0.03%-0.09%之间;总有机碳(TOC)含量最底层(D30,1.33%)明显高于其他层(0.43%-0.60%);而TOC:TN比值则是20 cm以下(10.11-15.61)明显高于20 cm以上(7.23-7.78)。沉积物中溶解性无机氮(dissolved inorganic nitrogen,DIN)以氨氮(NH4+)为主,NH4+浓度在5 cm和15 cm深度较高,达到1.64-2.34 mg/L,20 cm以下非常低,仅为0.10-0.22 mg/L。在25 cm深度以上,沉积物粒径较小(< 13 μm),25 cm和30 cm深度粒径超过34 μm。金属离子中Fe、Mn、Pb、V、Cr、Cu、Zn和As含量相对较高,这些金属离子在不同沉积物深度间差别不大。

2.2 细菌和古菌16S rRNA基因的拷贝数

荧光定量PCR结果(图 1)显示,细菌16S rRNA基因拷贝数范围为(0.09-14.45)×108 copies/g,古菌拷贝数范围为(0.89-20.59)×106 copies/g,前者比后者高出2个数量级。在垂直剖面上,古菌和细菌拷贝数呈现出一致的变化趋势,即表层(5 cm)较高层和中层(15 cm和20 cm)降低、25 cm层有所回升、30 cm层又下降的趋势。不同深度的ANOVA检验发现,细菌16S rRNA基因拷贝数在5 cm层显著高于20 cm层和30 cm层(P < 0.05),古菌16S rRNA基因拷贝数在不同深度间差异不显著(P > 0.05)。

2.3 不同深度沉积物细菌和古菌的α多样性

通过质控共获得1 331 066条高质量的细菌16S rRNA基因序列,划分为18 150个OTU,各层间的OTU数量平均在3 583-4 122之间;由于部分样品(column 1的D20、D25、D30,column 2的D10、D15、D20,column 4的D20)古菌V4区的扩增量无法满足高通量测序要求,因此只获得来自其他16个样品的633 307条古菌16S rRNA基因序列,划分为3 090个OTU,各层间OTU数量平均为425-657个。

细菌的α多样性指数(Chao1、OTU richness、Simpson和Shannon)在最表层(5 cm)呈现最大值,随着深度增加,均有降低的趋势,但在25 cm和30 cm层,α多样性指数反而比中层(15 cm和20 cm)略高,呈现“V”形分布(表 2)。古菌的α多样性指数最高值不在最表层(5 cm),而在次表层(10 cm);随着深度增加,α多样性指数均呈减低趋势,在30 cm层,α多样性指数最低。通过ANOVA检验,沉积物深度对海草床沉积物细菌和古菌的α多样性指数无显著影响(P > 0.05),各层间也无显著差异(P > 0.05) (表 2)。

2.4 海草床柱状沉积物中的细菌和古菌群落组成

图 2A可以看出,在所有柱状沉积物的细菌中,变形杆菌门(Proteobacteria)占据绝对优势(52.38%±1.83%),其次是绿弯菌门(Chloroflexi,14.27%±1.35%)、拟杆菌门(Bacteroidetes,7.88%±0.53%)、浮霉菌门(Planctomycetes,4.09%±0.34%)、放线菌(Actinobacteria,3.08%±0.20%)和其他一些低丰度细菌门。变形菌中,δ-变形菌纲(Deltaproteobacteria,27.93%±0.92%)丰度最高,其次是γ-变形菌纲(Gammaproteobacteria,12.71%±0.95%)、α-变形菌纲(Alphaproteobacteria,7.43%±0.46%)、ε-变形菌纲(Epsilonproteobacteria,3.3%±1.41%)和β-变形菌纲(Betaproteobacteria,0.55%±0.04%)。绿弯菌门主要由厌氧绳菌纲(Anaerolineae,8.36%±0.75%)和脱卤拟球菌纲(Dehalococcoidia,3.43%±0.51%)组成。拟杆菌门主要由黄杆菌纲(Flavobacteriia,3.47%±0.30%)组成。

在古菌中(图 2B),深古菌门(Bathyarchaeota)是第一大优势古菌门(46.99%±4.86%),其次是乌斯菌门(Woesearchaeota,21.29%±4.64%)、洛基古菌门(Lokiarchaeota,8.13%±1.03%)、Marine hydrothermal vent group (MHVG,7.17%±0.86%)、广古菌门(Euryarchaeota,6.84%±0.87%)、奇古菌门(Thaumarchaeota,6.20%±1.09%)和其他一些相对丰度小于1%的古菌门。深古菌门的子类群中相对丰度最高的是Bathy_17 (13.45%±1.54%),然后是Bathy_1 (8.28%±3.36%)、Bathy_6 (6.32%±0.99%)、Bathy_8 (5.54%±0.67%)、Bathy_10 (4.57%±1.40%)和Bathy_Other (2.84%±0.37%)。乌斯菌门的主要类群有Woese_5b (13.07%±4.36%)、Woese_5a (2.32%±0.43%)和Woese_Other (1.85%±0.33%)。

2.5 海草床沉积物细菌和古菌的垂直分布

nMDS结果显示(图 3),细菌和古菌的群落组成在不同深度沉积物间均未分开。进一步的ANOSIM检验发现(表 3),基于Bray-Curtis距离(R=0.02,P=0.40)和种间进化关系的Weighted UniFrac距离(R=0.06,P=0.20)分析表明,沉积物深度对细菌的群落分布均无显著影响。层间比较发现,5、10和15 cm深度间差异显著(P=0.03)。沉积物深度对古菌的群落分布无显著影响(P > 0.05),而且不同层间的古菌群落结构也无明显差异(P > 0.05)。

2.6 细菌和古菌主要类群在不同深度间的差异

通过ANOVA比较发现(表 4),细菌中最优势类群变形菌门的相对丰度在20 cm层(42.4±4.1%),显著低于其他层(高于50%) (P < 0.05),其中δ变形菌纲在最底层丰度最高(31.79±0.86%),15 cm到25 cm层的丰度较低(22.93-26.8%) (P=0.01)。拟杆菌门的垂直分布趋势与变形菌门相似。绿弯菌门的分布趋势则相反,20 cm层(22.6±2.6%)显著高于其他层(10%-15%左右) (P < 0.05);浮霉菌门的相对丰度则是中、下层明显高于表层(P=0.04)。蓝细菌门和酸杆菌门分别在10 cm和15 cm处相对丰度较高。其他类群层间差异不显著(P > 0.05)。

从总体上看(表 5),古菌中只有奇古菌门在沉积物各层间差异显著(ANOVA,P=0.01),其中主要类群是Soil crenarchaeotic group (SCG),最高丰度在15 cm层(11.6%±2.8%),最低丰度在30 cm层(0.7%±0.3%)。最优势的深古菌门在25 cm层相对丰度最高(70.2%±10.9%),10 cm层相对丰度最低(36.5%±5.0%)。其中Bathy_6类群在15 cm层丰度(11.9%±4.0%)显著高于最底层(3.4%±1.4%) (P < 0.05),前者是后者的3倍多;Bathy_10类群在25 cm层的丰度(11.8%±6.1%)显著高于表层(5 cm,4.5%±2.7%)、次表层(10 cm,1.4%±0.3%)和20 cm层(1.4%±0.0%) (P < 0.05)。其他类群如Bathy_17、Bathy_8和Bathy_13的最小丰度均在底层,而类群Bathy_1则是底层较高,相对丰度约为表层和中层的4-10倍。第二大类群乌斯菌门在表层(27.5%±11.1%)和底层(30.0%±14.6%)丰度较高,中层(15-25 cm)较低(6.5%-8.8%);而广古菌门则呈现了相反的趋势,相对丰度中层最高。

3 讨论 3.1 深度对海草床沉积物理化性质的影响

海草生长的过程中,凋落物不断在沉积物中累积,形成巨大的碳汇,因此海草床沉积物通常有机质含量较高[25]。海草床沉积物不同深度有机质的组成差别较大,深层主要由难降解的惰性有机质如纤维素、半纤维素、木质素和其他大分子的有机质组成,而浅层沉积物中除了这些惰性有机质,还有大量不稳定的活性有机质,其来源于海草根际分泌的光合作用产物,以及水体中浮游和附生藻类等的残片[25-26]。碳氮比是指征有机质活性程度的重要指标[27-28],碳氮比低,说明有机质含氮物质高,易氧化和被微生物分解,多为活性物质;而碳氮比高,说明大分子含碳物质(如木质素)含量高,不易降解,多为惰性物质。在本研究中,表、中层(D5-D15)沉积物的TOC:TN比值在7.5左右,远低于深层(D20-D30)沉积物的比值(10-15) (表 1),说明该海草床沉积物的有机质组成确实存在明显的垂直特征,即浅层富含活性有机质、深层富含惰性有机质。微生物会迅速利用这些不稳定的活性有机物。而且,这些活性有机质还可能通过“激发效应”(priming effect)[29],即刺激微生物的生长,从而提高胞外酶的分泌,加速惰性有机碳的降解。这就不难理解我们海草床浅层沉积物的有机碳含量反而低于深层的现象;另外,由于浅层活性有机质分解产生大量NH4+,因此浅层沉积物NH4+含量明显高于深层。

3.2 深度对海草床沉积物微生物丰度的影响

研究发现,海草的根和根状茎大部分(90%以上)集中在沉积物40 cm以上的深度[30-31]。在垂直剖面上,海草根生物量(root biomass)一般在10 cm左右出现峰值,然后随深度的增加而递减[32]。我们通过荧光定量PCR技术发现,细菌和古菌的16S rRNA基因拷贝数也是在10 cm以上较高,然后随深度增加逐渐下降,可见沉积物垂直剖面的微生物丰度与海草根生物量关系密切。这可能是受根组织分泌的有机质在垂直方向上的梯度驱动[32]。事实上,在很多沉积物和土壤研究中,微生物丰度都是在表层最高,然后随深度增加迅速下降[16, 33]。值得注意的是,在我们的结果中,不论细菌还是古菌16S rRNA基因拷贝数在25 cm层都出现一个峰值,这与红树林沉积物中一些微生物类群的最大丰度不在表层而出现在20-25 cm层一致[15]。推测这些类群可能是厌氧类群,而且擅长降解利用大分子惰性有机质。

3.3 海草床沉积物细菌群落结构的垂直特征

在各种深度的海草床沉积物中,变形菌门(Proteobacteria)均占有绝对优势,特别是δ变形菌纲,其相对丰度约为细菌组成的1/3,这与红树林沉积物中δ变形菌纲的比例相似[34]。δ变形菌纲中的很多类群属于硫酸盐还原菌,这些类群能够利用多种有机质作为电子供体还原硫酸盐。据报道,在海草床系统中硫酸盐还原菌负责超过50%的有机质氧化[35]。绿弯菌门(Chloroflexi)也经常出现在各种有机质丰富的土壤和沉积物生境中[36],其中的厌氧绳菌纲(Anaerolineae)是严格厌氧菌,经基因组测序发现,厌氧绳菌具有降解纤维素的功能[37],该类群在20 cm层相对丰度较高。拟杆菌门(Bacteroidetes)中的黄杆菌纲(Flavobacteriia)是重要的海洋异养菌类群,在海水食物链和碳循环中扮演重要角色[38-39],特别擅长大分子有机质如纤维素、木聚糖和几丁质的降解[40],该类群在表层和底层丰度较高,而中层丰度较低。浮霉菌门(Planctomycetes)的比例在20 cm以下明显升高,研究发现[24, 41]该类群是降解高分子量化合物和难降解物质的关键细菌。不难看出,能够利用多种有机质的变形菌、拟杆菌较偏爱表层环境,而随着深度的增加,易降解的活性有机质减少,惰性有机质比重增加,细菌类群对碳底物的需求具有一定的特异性,具有降解大分子有机质能力的类群逐渐占优势。海草床沉积物细菌的这种垂直分布特征与富含有机质的河口沉积物中细菌的垂直分布特征非常相似[42]

3.4 海草床沉积物古菌群落结构的垂直特征

深古菌门(Bathyarchaeota)是海草床沉积物中最优势的古菌,相对丰度接近50%,这与其在红树林沉积物中的比列相似[16],而且不论在海草床还是红树林的沉积物中,该类群均在25 cm左右比例很高。深古菌门可利用多种形式的有机质,如糖类和蛋白质碎屑等[33]。其中Bathy_8可以利用木质素[43],而海草的根、茎中存在大量木质素[10, 44],这些信息暗示了该类群可能在降解海草根茎残片中起重要作用。类群Bathy_6的相对丰度在15 cm最高,然后随着深度加深而迅速降低,这是因为Bathy_6偏爱低硫化物浅层沉积物生境[15],由于海草床沉积物富含硫酸盐和有机质,在厌氧层硫酸盐还原菌利用有机质还原硫酸盐产生大量H2S,抑制了Bathy_6的生长,而浅层根输入O2营造的低还原环境则有利于该类群的生存。

作为海草床沉积物中第二大优势古菌,乌斯菌门(Woesearchaeota)在各层间的平均相对丰度在20%左右,主要集中在表层和底层,而中层(15-25 cm)较低。目前,虽然该类群经常在生产力较高的环境中被发现且比例很高[45-46],然而其在环境中的生态功能还不清楚。根据前人的研究[45, 47],该类群可能在厌氧碳循环中发挥重要作用。基于宏基因组分析[45],乌斯菌门具有在厌氧条件下发酵醋酸盐和产H2的潜能,而且由于缺失核心的生物合成途径,推测其可能营共生和/或发酵的生活方式[45, 47]。网络分析显示,乌斯菌在环境中与甲烷菌高度共存[45]。因此我们推测,在表层沉积物中,由于活性有机质较多,其他微生物类群代谢活跃,乌斯菌可利用其他类群的代谢副产物生存;而在深层厌氧沉积物中,乌斯菌可能与甲烷菌协同共生,为甲烷菌提供H2和醋酸盐等底物,并利用甲烷菌的代谢产物生存[45, 47]

奇古菌门(Thaumarchaeota)相对丰度在15 cm以下随深度增加而迅速降低,这是因为该类群中很大比例属于氨氧化古菌[48],其对海洋NH4+氧化具有重要贡献,NH4+氧化过程需要O2参与,因此随着深度的增加、根茎组织的减少,沉积物中的O2水平迅速降低,氨氧化古菌受抑制;另一方面,随着深度增加,NH4+浓度也迅速降低,氨氧化古菌会因为底物限制而受抑制。

4 结论

基于荧光定量PCR和高通量测序技术,我们分析了海草床沉积物0-30 cm剖面细菌和古菌绝对丰度(拷贝数)、多样性和群落结构的垂直特征。结果发现,细菌和古菌的丰度呈现出随沉积物深度增加而降低的趋势,25 cm深度有所回升然后又下降。虽然细菌和古菌的整体群落组成在沉积物不同深度间差异不显著,但某些类群表现出明显的层间差异,推测可能受沉积物中有机质组成、溶氧状态和氮含量的影响。我们的研究从丰度和多样性角度阐述了细菌和古菌在海草床沉积物不同深度可能发挥的生态功能和环境响应,进一步的研究应从宏基因组、胞外酶和底物利用等层面进一步证实这些功能。
