我国南北区域城市污水处理系统内真菌群落的差异

摘要: 【背景】 微生物是污水处理系统内污染物去除的主体,真菌作为其中不可或缺的一部分,在去除有毒化合物、提高生物转化等方面发挥着重要作用,应引起人们的关注。【目的】 明确我国南北区域城市污水处理系统内真菌群落的多样性、组成差异及其影响因素。【方法】 采集我国北方23个污水处理厂的90份活性污泥样品和南方37个污水处理厂的121份活性污泥样品,提取上述样本的总DNA,并应用Illumina MiSeq平台进行ITS高通量测序,结合多种数量生态学分析方法对数据进行分析。【结果】 我国南北区域城市污水处理系统内真菌群落结构存在显著差异,真菌群落多样性南方显著高于北方;我国南北区域真菌群落的组成差异显著,南方群落以SordariomycetesGlomeromycetes为优势菌纲,以OphiocordycepAlternaria为优势菌属,北方群落以TremellomycetesSaccharomycetes为优势菌纲,以TrichosporonSaccharomyces为优势菌属,其中TremellomycetesSordariomycetes是常见的病原菌,对下游生物或者人类的健康具有潜在威胁,Trichosporon属的丝状真菌若大量异常增殖会引发污泥膨胀现象,影响系统的稳定运行;地理纬度、年平均气温、进水NH4+浓度、进水总氮浓度是导致我国南北区域真菌群落存在差异的重要影响因子。【结论】 我国南北区菌群落多样性、组成存在显著差异,地理因素、所在城市的气候因素和进水污染物浓度对真菌群落结构影响显著。针对南北区域,应合理调控相应区域内的重要功能菌群,以调控污水处理厂的高效稳定运行,同时密切关注其中的条件致病菌和易引发污泥膨胀的菌群,建立面向种群的优化控制系统以管控风险。

Differences of fungal community in wastewater treatment plants between south and north China

Abstract: [Background] Microorganisms are main agents to remove pollutants in activated sludge (AS) in wastewater treatment plants (WWTPs), among them, fungi should be paid attention to as they play important roles in removing toxic compounds and improving biological transformation, etc. [Objective] The major research will be focus on to characterize and compare the diversity, composition of fungal communities in WWTPs located in north and south China, determine the influence factors of fungal community structure. [Methods] Ninety AS samples from 23 WWTPs in north China and 121 AS samples from 37 WWTPs in south China were collected, respectively. After DNA extraction and amplification, high-throughput sequencing of ITS gene was carried out using Illumina MiSeq platform. Data were processed by a variety of quantitative ecological analysis methods. [Results] Fungal community structures in AS system between north and south China were significantly different. Diversity of fungal community in south China was higher than that of north China. The dominant classes and genera in fungal community were Sordariomycetes and Glomeromycetes classes, and Ophiocordycep and Alternaria genera in south China. While they were Tremellomycetes and Saccharomycetes classes, and Trichosporon and Saccharomyces genera in north China. Members of the classes Tremellomycetes and Sodariomycetes are opportunistic pathogens, which may be potential threats to downstream organisms or human health. Abnormal growth of members in Trichosporon genus will cause sludge bulking and affect the stable operation of WWTPs. Latitude, annual average air temperature, concentration of NH4+ and the total nitrogen in influent were identified as the important factors that influence fungal community structure in WWTPs. [Conclusion] Fungal communities between north and south China were significantly different in diversity and composition as a results of geographic factors, climate factors and concentration of pollutants in influent. Accordingly, toward the efficient and stable operation of the treatment plants, it is necessary to regulate the different important communities in south and north China. Furthermore, we should pay close attention to the opportunistic pathogenic fungi and some agents that easily cause sludge bulking. Establishment of population-oriented optimal control system is necessary to control the risks.

城市污水处理厂(Wastewater treatment plant,WWTP)是人类和城市发展的重要设施,在污水生物处理过程中,微生物是污染物去除的主要承担者,因此微生物群落与系统的稳定运行密切相关。近年来相关研究逐渐揭示出真菌在污水处理系统内的重要作用,如促进硝化作用和反硝化作用[1]、去除有毒化合物、促进生物絮凝[2]、脂质积累[3]、提高生物转化[4]和脱水性能[5]等。但是,丝状真菌若大量异常增殖,则会引发污泥膨胀现象,严重影响处理系统的稳定运行[6]。因此,深入理解并合理利用污水处理系统中的真菌群落,对保障系统稳定运行具有重要作用。

在污水处理系统微生物群落中,真菌的多样性大于古菌和其他一些真核微生物[3],但是真菌的相关研究却很少。过去的很长一段时间内,活性污泥内真菌群落的研究依靠传统的微生物培养方法和形态学研究手段[1-2],近年来高通量测序技术的应用极大地扩展了我们对真菌群落的认识,但是相关研究多集中于膜生物反应器(Membrane bio-reactor,>MBR)和生物滤池等工艺内的真菌群落[7-9],大规模区域尺度的污水处理系统真菌群落的研究工作尚未开展。此外,我国幅员辽阔,地域差异显著,自然、人文条件各异,各地区居民的饮食结构多样,有研究显示自然系统内真菌群落存在着显著地域性差异[10],真菌群落的组成相似性随地理距离的增加而减小,即符合距离-衰减规律[11]。污水处理厂作为工程构筑物,活性污泥系统内细菌群落也存在着地域性差异[12],也符合距离-衰减规律[13],但是不同区域活性污泥系统内真菌群落的差异仍属未知。

因此,本研究利用高通量测序技术,结合多种生态学及数理统计相关分析方法,解析我国南北区域城市污水处理厂活性污泥系统中真菌群落的基本特性及影响因素,以期深入认识污水处理厂活性污泥系统内的真菌群落,丰富与拓展现有生态学知识与理论,为我国不同区域污水处理厂建立面向种群的优化控制系统提供技术支撑。

1 材料与方法 1.1 主要试剂和仪器

PowerSoil® DNA提取试剂盒,MoBio公司;DNA纯化试剂盒MinElute® Gel Extraction Kit,QIAGEN公司。多参数水质测定仪,Thermo Scientific公司;PCR仪、凝胶成像系统,Bio-Rad公司。

1.2 样品采集与DNA提取

2014年夏季,采用统一方法对我国15个城市的污水处理厂进行了样品采集。根据地理环境的差异将我国划分为南北两大区域,南方区域包括9个城市,即无锡、上海、武汉、长沙、成都、重庆、厦门、深圳和三亚;北方区域包括6个城市,即哈尔滨、大连、北京、济南、青岛和西安。共采集城市污水处理厂内活性污泥样品211份,其中南方样品121份,北方样品90份,涉及南方37个污水处理厂和北方23个污水处理厂。采样时,南方各城市的年平均气温为17.22-25.56 ℃,北方各城市的年平均气温为4.44-13.89 ℃,具体样品采集原则、方法参考张冰等[14]。同时,收集污水处理厂的地理信息、所在城市的气候信息、WWTPs的设计参数、进出水水质情况和操作参数等基本信息,原位测定曝气池混合液的温度、溶解氧浓度和pH等参数。

利用PowerSoil® DNA提取试剂盒对离心后的污泥样品进行DNA提取,然后利用分光光度计测定DNA浓度和纯度,以满足测序的DNA质量要求。

1.3 ITS高通量测序

利用引物ITS7F (5′-GTGARTCATCGARTCTT TG-3′)和ITS4R (5′-TCCTCCGCTTATTGATATGC-3′)[15]对满足要求的样品DNA进行两步法PCR扩增[16],产物纯化后通过Illumina MiSeq测序平台进行ITS测序,然后对合格的测序数据进行整合和物种信息注释分析,继而生成操作分类单元(Operational taxonomic unit,OTU)表格,数据预处理过程详见文献[17-18]。

1.4 数据分析

采用非度量多维尺度法(Nonmetric multidimensional scaling,NMDS)和相异性分析非参数检验方法,包括置换多变量方差分析(Permutational multivariate analysis of variance,PERMANOVA,又称ADONIS)、相似性分析(Analysis of similarities,ANOSIM)和多响应置换过程(Multi-response permutation procedure,MRPP),检验我国南北区域真菌群落结构的差异;利用R计算多样性指数[19];利用线性判别分析方法(Linear discriminant analysis effect size,LEfSe)解析我国南北区域真菌群落组成差异[20],设置线性判别分析(Linear discriminant analysis,LDA)筛选值为3.0;分别采用Partial mantel test和典范对应分析(Canonical correspondence analysis,CCA)方法确定影响真菌群落结构的因素。以上分析通过Galaxy平台[21]、R语言的vegan、ape、ecodist程序包完成。

2 结果与分析

为了保证样品间数据的均等性,按照每个样品9 389条序列的标准对测序结果进行随机抽平处理,211个样品共得到1 981 079条序列,然后按照97%相似度的标准对序列进行OTU划分,211个样品共获得了12 925个OTU,其中南方真菌群落121个样品共获得了10 310个OTU,北方真菌群落90个样品共获得7 465个OTU,二者共有4 850个OTU,占OTU总数量的37.5%,其中南方特有OTU为5 460个,占OTU总数量的42.2%,北方特有OTU为2 615个,占OTU总数量的20.2%。

2.1 我国南北区域污水处理厂活性污泥系统内真菌群落多样性的差异

2.1.1 β多样性

采用基于Bray-Curtis距离的NMDS方法对我国污水处理厂活性污泥系统内真菌群落进行了分析,结果显示,整体上,来自南、北方污水处理厂的活性污泥样品各自聚为一类(图 1),表明我国南北区域的活性污泥系统内真菌群落结构具有显著差异。利用MRPP、ADONIS和ANOSIM对我国南北区域的活性污泥系统内真菌群落结构进行相异性分析,结果显示,南北区域真菌群落的确存在显著差异(P < 0.01),相异性分析结果佐证了上述结论。

2.1.2 α多样性

α多样性指数是表征微生物群落内物种丰富度和均匀度的重要参数。我国南方区域污水处理厂活性污泥系统内每个样品中真菌群落的OTU数为162-1 237,估测丰富度Chao1指数为313- 1 690,香农-威纳指数为1.11-5.52,均匀度指数为0.20-0.78;我国北方区域活性污泥系统内每个样品中真菌群落的OTU数为131-847,估算的OTU丰富度Chao1指数为231-1 621,香农-威纳指数为0.61-5.10,均匀度指数为0.12-0.78。整体上,我国南方真菌群落物种丰富度和估测丰富度(Chao1)均显著高于北方(图 2),南方真菌群落的香农-威纳指数和均匀度也高于北方。

2.2 我国南北区域污水处理厂活性污泥系统内真菌群落组成的差异

为了更好地探究真菌群落的地域性组成差异,采用LEfSe方法对我国南北区域真菌群落进行了差异分析,结果显示我国南北区域真菌群落组成差异显著(图 3A),当LDA值大于3.0时,共得到54个生物标记物(Biomarker) (图 3B),其中北方真菌群落的生物标记物40个,南方真菌群落的生物标记物14个,这些生物标记物在南北区域真菌群落内的相对丰度差异显著。

具体地,北方真菌群落内LDA值最大的是Tremellomycetes,南方群落内LDA值最大的是Ophiocordyceps,说明二者在我国南北区域真菌群落内的相对丰度差异最大。纲水平上,SordariomycetesGlomeromycetes在南方真菌群落内的相对丰度显著高于北方,TremellomycetesSaccharomycetesDothideomycetesEurotiomycetesPezizomycetes在北方真菌群落内的相对丰度显著高于南方。属水平上,OphiocordycepAlternariaSebacina在南方真菌群落内的相对丰度显著高于北方,TrichosporonSaccharomycesPseudeurotiumPichiaEmmonsiaPenicilliumMucorAcidomycesScutelliniaInocybeSynchytriumCephalotheca在北方真菌群落内的相对丰度显著高于南方。

2.3 我国污水处理厂活性污泥系统内真菌群落结构的影响因素

为了探明影响我国污水处理厂活性污泥系统内真菌群落结构的影响因素,利用Partial mantel test对污水处理厂的地理信息、所在城市的气候信息、进出水水质信息、操作条件等参数与群落结构的相关性进行了分析,图 4结果表明,在参与分析的15个因子中,5个因子与活性污泥系统内真菌群落结构显著相关(P < 0.05),其中相关系数最大、相关性最强的因子是地理纬度(r=0.210,P=0.001),其次是年平均气温(r=0.185,P=0.001)和曝气池混合液浓度(r=0.166,P=0.003),进水污染物浓度与群落结构存在很强相关性,其中进水NH4+浓度(r=0.123,P=0.004)和进水总氮浓度(r=0.126,P=0.009)与群落结构相关性较强,进水COD浓度、进水BOD浓度和进水总磷浓度与群落结构则没有显著相关性。此外,操作条件中仅污泥龄(r=0.091,P=0.048)与群落结构存在显著相关性,曝气池溶解氧浓度、水力停留时间等操作条件与活性污泥系统内真菌群落结构则不存在显著相关性。

同时,利用基于单峰模型的CCA分析污水处理厂地理信息、所在城市气候信息、进出水水质信息、操作条件等参数对群落结构的影响,结果表明,地理信息(地理纬度、经度、年平均气温)、进水水质信息(进水NH4+浓度、进水总氮浓度和进水总磷浓度)和曝气池混合液浓度、温度、溶解氧浓度、水力停留时间、污泥龄等因素对全国污水处理厂真菌群落结构均有不同程度的影响,共解释群落结构变化的35.88%,其中第一轴(CCA1)占总解释量的12.63%,第二轴(CCA2)占总解释量的11.68%。综合考虑各因素在坐标轴上的投影长度和与坐标轴的夹角,地理纬度、经度及年平均气温、进水NH4+浓度、进水总氮浓度、进水总磷浓度、水力停留时间与群落结构具有较强相关性,对我国污水处理厂真菌群落结构的影响较大。

由于CCA和Partial mantel test所基于的算法和模型的差异,分析结果稍有差异,综合考虑后将两种分析方法结果中均与群落显著相关的环境因子作为重要环境因子,因此,对全国污水处理厂真菌群落结构影响较大的因子是地理纬度、年平均气温、进水NH4+浓度、进水总氮浓度。

3 讨论与结论

在污水处理系统中,真菌的作用不可忽视,然而相关研究较少,且存在研究手段落后、尺度小、认识不足等问题,因此,本研究利用高通量测序技术并结合多种生态学及数理统计相关分析方法,解析我国南北区域城市污水处理厂活性污泥系统中真菌群落的基本特性及影响因素,以期深入认识污水处理厂活性污泥系统内的真菌群落,丰富与拓展现有生态学知识与理论。

比较分析我国南北区域活性污泥系统内真菌群落多样性发现,我国南北区域真菌群落结构存在显著差异,这与自然系统内真菌群落的地理分布规律一致,Wu等[10]分别采用分离培养和聚合酶链反应-变性梯度凝胶电泳的方法,评估了中国10个长江沿岸湿地和其他10个地区湿地沉积物中的真菌群落,结果显示,20个采样点的样品共分为三类:中国南方、中国北方和青藏高原,说明真菌群落存在显著的地域性差异。

与城市供水系统内真菌群落相比,污水处理系统内真菌群落的多样性较高,如王钰等[22]研究城市供水系统中真菌群落多样性,结果显示单个样品内的香农-维纳指数为1.87-4.34,Chao1指数为44-391个OTU。与活性污泥系统内的细菌群落比较发现,真菌群落物种多样性较低,如张冰等[14]对全国污水处理厂活性污泥系统内细菌群落分析发现,211个样品共获得了33 849个OTU;Xia等[23]利用Illumina测序方法分别对MBR和氧化沟(Oxidation ditch,OD)系统内12周的活性污泥样品进行了分析,结果表明MBR和OD系统样品的OTU数目分别为2 297-3 330个和1 802-3 517个,远高于真菌群落多样性;Zhang等[12]利用454焦磷酸测序技术获得活性污泥系统内细菌群落OTU数目为1 183-3 567个。Wei等[24]对比分析了城市污水处理厂内真菌和细菌的季节和空间的动态变化,获得5 756个细菌OTU,2 469个真菌OTU,细菌的香农-威纳指数为5.32-5.73,远高于真菌的香农-威纳指数1.49-5.05。

与森林土壤系统内的真菌群落多样性相比,污水处理系统内的真菌群落物种丰富度和多样性较低,如Buée等[25]利用基于ITS的454焦磷酸测序技术评价了6个不同森林土壤内真菌群落多样性,结果显示每个样品内的物种丰富度约为1 000个OTU;Wubet等[26]利用基于ITS的焦磷酸测序技术分析了欧洲以山毛榉占主导地位的森林土壤真菌多样性和群落组成,结果显示,每个样品内真菌群落丰富度为1 655个OTU。然而,与耕作或草原土壤系统内的真菌群落多样性相比,污水处理系统内的真菌群落物种丰富度和多样性较高。Klaubauf等[27]利用基于ITS/LSU区域的RFLP分析了奥地利的4种耕作土壤和1种草原土壤真菌多样性,结果显示5种土壤中真菌群落丰富度为19-34个OTU。Moll等[28]利用基于ITS/LSU区域和SSU区域的克隆技术,结合Sanger测序方法研究玉米田土壤内普通真菌和丛枝菌根真菌群落的特性及空间分布,结果显示真菌群落的丰富度仅为313个OTU。此外,活性污泥系统真菌群落丰富度远高于入湖河口沉积物和街道空气内的真菌群落丰富度,如王鹏等[29]利用18S rRNA基因高通量测序技术分析了鄱阳湖入湖河口14处沉积物的真菌群落结构特征,结果显示单个样品内真菌群落物种丰富度为62–194个OTU;王琳等[30]通过构建18S rRNA基因克隆文库方法分析青岛市市区街道秋季空气微生物群落多样性,结果显示空气真菌群落丰富度为17个OTU。

本研究利用线性判别分析方法解析了我国南北区域真菌群落的组成差异,结果显示,南方群落中SordariomycetesGlomeromycetes为优势菌纲,OphiocordycepAlternaria为优势菌属,北方群落中TremellomycetesSaccharomycetes为优势菌纲,TrichosporonSaccharomyces为优势菌属。据文献报道,能将NO2还原为N2O的真菌大多隶属于子囊菌门的SordariomycetesEurotiomycetes,它们极大地促进了废水中氮的去除[31]TremellomycetesBasidiomycotaTremella分支的一个主要成员,是活性污泥系统内的机会动物病原菌[32-33];同样地,Sordariomycetes是常见的污水处理厂和养殖池塘内的动物病原菌[34],这些条件致病菌的存在可能对下游生物或者人类的健康造成威胁,因此应重点关注。同时,Trichosporon属的丝状真菌若大量异常增殖会引发污泥膨胀现象,造成水质下降和污泥的大量流失,影响处理系统的稳定运行,因此北方污水处理厂在运行过程中应着重关注该属的动态变化。此外,活性污泥中的Trichosporon属成员在污水处理过程中具有重要的作用,如T. cutaneum能够去除COD,降解芳香族化合物[35-36]Saccharomyces cerevisiae[37]Candida lipolytica[38]能够吸收重金属,因此,在污水处理厂运行过程中,或可通过合理利用这些功能菌群建立面向种群的优化控制系统,调控污水处理厂的高效稳定运行,同时应密切关注条件致病菌和易引发污泥膨胀的菌群,管控风险。

基于生态位理论描述的确定性因素[39]和基于中性理论的随机因素[40]是目前公认的影响微生物群落多样性和结构的因素,现有研究多关注确定性因素中的污水处理厂地理位置、水质条件和操作参数等因素对活性污泥系统内细菌群落结构的影响[41-43],也有研究结果表明,研究对象不同,影响作用较显著的因素种类也可能存在差异[44],然而对于真菌群落影响因素的探究较少,且结论尚不明确。为了进一步明确影响我国南北区域真菌群落结构与组成差异的因素,本研究分别利用Partial mantel test和CCA两种方法对我国污水处理厂内真菌群落进行了分析,结果显示,地理纬度、年平均气温、进水NH4+浓度、进水总氮浓度对我国污水处理厂内真菌群落结构的影响较大。

与污水处理厂内细菌群落类似,真菌种群的变化与环境变量、操作参数的变化具有很强的相关性。Wei等[24]分析了影响城市污水处理厂活性污泥系统内的细菌和真菌群落的因素,结果表明,地理距离、曝气池溶解氧浓度、气温、出水总氮浓度、水力停留时间、污泥龄、进水COD浓度与微生物群落结构均存在较强的相关性。Maza-Márquez等[45]利用18S rRNA基因和ITS作为分子标记,并结合温度梯度凝胶电泳和454焦磷酸测序对MBR反应器不同运行阶段内真菌群落进行了全面监测,结果显示,真菌种群的变化与环境变量和操作参数变化确实具有很强的相关性,其中pH的相关性最强。此外,相关研究表明,地理距离的差异导致了巨大的环境异质性,如气温、降水量,进而导致生态系统中微生物种群的地域性分化[46],本研究中Partial mantel test和CCA两种分析方法均表明地理纬度对我国污水处理系统内真菌群落结构有重要影响。

与污水处理厂内细菌群落相比,真菌群落研究成果较少且数据库不全面,因此很多物种分类地位不明确,这也大大增加了研究的难度,限制了研究的深度和广度,相信随着基于高通量技术的微生物基因组学的发展,污水处理系统内真菌群落的“黑箱”将逐渐被打开。
