石油等化石燃料是目前国际上的主要能源物质,但由于其储量的不断下降、人类对能源需求的持续增长以及世界各国对环境保护意识的不断增加[1-2],寻找可替代化石能源的生物质能源成为近年来能源领域的研究热点[3]。生物丁醇作为第二代能源物质是一种全新的、可再生的绿色生物能源,且具备替代化石能源的潜力,因此生物法获取丁醇重新燃起了研究者的兴趣[4-5]。
至今,丙酮丁醇(Acetone-butanol-ethanol,ABE)发酵研究已有150多年历史,在20世纪60年代前发酵法生产丁醇是仅次生物乙醇的第二大发酵工业[6]。然而,传统ABE发酵工业通常是以玉米粉和糖蜜等食品原料进行发酵生产[7-8],与石油化工衍生生产丁醇相比,因其具有原料成本高、丁醇浓度低等缺点,导致发酵法生产丁醇在经济上没有竞争力,美国于20世纪60年代初停止发酵法生产丁醇[9-10]。
虽然丁醇发酵研究已有150多年历史,但利用以木质纤维素基质为原料的研究在近年才被广泛开展。目前,ABE发酵工业采用的生产菌种主要为梭菌属,其主要优点是具有将葡萄糖、蔗糖、木糖、乳糖和淀粉(玉米粉、木薯粉)等多种底物进行生物转化的能力[11-12]。由于木质纤维素由各种单糖组成且是地球上最为丰富的可再生资源,使其在生物燃料生产中成为可持续和具有成本效益的生物质[13]。因此,在丁醇生产过程中从木质纤维素生物质中获取可发酵糖是最经济有效的丁醇生产策略之一[14]。到目前为止,已被成功研究用于ABE发酵的木质纤维素材料有:麦杆[15]、糖枫木[16]、玉米芯[17]、柳枝[18]、稻杆[19]、桉树[20]、麦麸[21]、玉米秸秆[22]和甘蔗渣[23]等,且利用这些材料进行ABE发酵均获得较高浓度丁醇。
本文综述了采用丙酮丁醇梭菌(Clostridium acetobutylicum)或拜氏梭菌(Clostridium beijerinckii)为生产菌株,并以木质纤维素生物质为原料生产丁醇可行性方面的研究进展,从木质纤维素生物质原料、木质纤维素生物质的预处理、抑制物对丁醇产生菌的影响、水解液脱毒及耐抑制物丁醇生产菌的选育等方面进行详细的综述,并对木质纤维素在生物燃料生产的应用前景进行了展望。
1 木质纤维素生物质 1.1 木质纤维素原料生物质能资源可分为4类:木制品工业废物、城市固体废物、农业残留物和专用能源作物。木质纤维素主要由纤维素、半纤维素和木质素等3种成分组成。部分木质纤维素原料中的纤维素、半纤维素和木质素含量如表 1所示[24-30]。半纤维素由一种不均一的分支多糖组成,主要由葡萄糖、木糖和其他五碳单糖组成,其平均分子质量小于30 000,纤维素是由β-1, 4-糖苷键将1, 4-D-葡糖基连接而成,其平均分子量约为100 000[28]。以木质纤维素生物质为原料与以玉米为原料发酵生产生物燃料相比,可以减少温室气体的排放[27]。因此,以木质纤维素生物质为原料进行生产生物丁醇有望解决ABE发酵过程中原料成本过高及减少其对环境造成污染等问题,从而提高ABE发酵的竞争力。

为使木质纤维素生物质所含单糖组成部分成为可发酵糖,需利用预处理方法将其组分中的刚性结构破坏并释放出可发酵糖。由于纤维素嵌入木质素和半纤维素相互结合的紧密复合结构中,并被一层半纤维素覆盖,使得原料预处理成为木质纤维素生物质生产丁醇的主要工艺挑战[31]。不同的预处理方法已被研究开发,但在工业规模上它仍然是第二昂贵的工艺[32]。然而,当廉价的木质纤维素生物质被分馏时,溶解的半纤维素作为发酵糖原料仍具有潜力。木质纤维素预处理方法可分为物理预处理法、化学预处理法、物理化学预处理法、生物预处理法等。
1.2.1 物理预处理常用的物理预处理方法有机械粉碎、微波处理、高能辐射及高温分解等。物理预处理可降低颗粒尺寸和结晶度、增加比表面积及降低聚合度以达到改善水解和增加纤维素酶与底物的结合,并不会破坏木质纤维素生物质的细胞壁结构,也几乎不改变半纤维素、纤维素和木质素的结构[28]。物理预处理的功耗相对较高,但取决于最终颗粒尺寸和生物质特性[32]。单独的物理预处理不能有效地完全水解生物质,往往需要与热化学预处理相结合。
1.2.2 化学预处理化学预处理已成为提高纤维素和半纤维素降解、去除木质素、降低木质纤维素中纤维组分的聚合度和结晶度的最有效的方法之一。化学预处理是指利用酸、碱、离子液体、氧化剂和有机溶剂等化学物质改变木质纤维素生物质的物理和化学特性[33]。目前,对木质纤维素丁醇生产的化学预处理方法已进行大量的研究[21, 34-37]。部分有机酸也可用作催化剂如:草酸、乙酰水杨酸和水杨酸等,且有机溶剂与无机酸的混合物也被用来破坏木质纤维素的刚性结构[38]。但化学预处理需要配备特殊的设备和仪器,如浓酸在水解过程中需要为其提供耐腐蚀性及耐高温高压的反应容器。
1.2.3 物理化学预处理物理化学预处理法可促进半纤维素溶解和改变木质素结构,也可促进纤维素酶与底物的结合[39-40]。由于其同时具有物理和化学预处理的性质,使其处理效果远高于物理预处理法。物理化学预处理法包括蒸汽爆破[41]、液体热水[42]、氨纤维/冷冻爆炸、湿式氧化、氨循环渗透、有机溶剂木质素和CO2爆炸等方法[43]。其中蒸汽爆破、液体热水等经常被用于对木质纤维素进行预处理发酵产生丁醇。
1.2.4 生物预处理生物预处理主要与能产生降解木质素、半纤维素的酶及多酚的白、棕、软腐真菌和某些细菌有关。由于生物预处理具有底物和反应特异性、低能量要求、不产生有毒化合物、产品高收率等潜在优点,从而引起了人们的广泛兴趣[44]。此外生物预处理还具有能量投入低、反应条件温和及环境友好型等优点。生物法的主要缺点是处理过程极其缓慢,有效的生物质水解需要几个星期到几个月的时间。因此,大规模应用生物预处理可能不是一种经济可行的生物燃料生产选择[45]。在木质纤维素生物质生产丁醇过程中纤维素酶常被用于水解经稀酸水解所剩残渣,以获得可发酵糖,从而提高木质纤维素生物质的转化效率。Yang等[13]利用纤维素酶对经稀硫酸水解残渣进行酶解,并在所获得的酶水解液中添加大麦淀粉浆,经ABE发酵后获得12.4 g/L的总溶剂。本实验室采用水磨预处理与酶(纤维素酶和木聚糖酶)糖化过程相偶联的方法对玉米芯进行水解,与单独水解法相比水解周期缩短58%,且经此法处理的还原糖浓度达到59.8 g/L,分别是酸处理法和碱处理法的1.3倍和1.7倍,以此未经脱毒的水解液为碳源,经优化发酵后丁醇产量达到9.5 g/L[46]。
2 抑制物对丁醇生产菌株的影响及水解液的脱毒 2.1 抑制物对产丁醇菌株的影响由于木质纤维素生物质结构紧密,导致从其中直接获得可发酵糖相当困难。虽然对木质纤维素生物质进行各种预处理过程可获得各种类的单糖,但也同时伴随出现各种抑制类化合物,这些物质会破坏细胞膜和细胞结构单元,使细胞不能发挥正常功能,预处理后所得的水解液中所含的抑制物主要成分有呋喃衍生物、低分子量脂肪酸和酚类化合物。
2.1.1 呋喃衍生物呋喃类化合物主要有糠醛和5-羟甲基糠醛(5-Hydroxymethyl furfural,HMF),并广泛存在于木质纤维素水解液中。当发酵液中含有糠醛时,菌体在产酸时期氧化还原平衡受到干扰且编码磷酸转移酶系统、细胞运动和鞭毛蛋白(及其组合)基因的表达受到抑制,从而导致菌体过早的终止发酵。Zhang等[47]研究表明,当糠醛和HMF浓度在1−2 g/L下时,糠醛和HMF对C. acetobutylicum的生长有短暂的抑制作用,导致滞后期延长,并当糠醛和HMF被还原为糠醇和2, 5-二羟甲基呋喃等毒性较小的抑制性化合物时,发酵才不会受到抑制。但在木质纤维素水解液中糠醛和HMF中的浓度往往较高,且大多数研究表明当呋喃类物质浓度高于2 g/L时会抑制丁醇的代谢[48]。高浓度呋喃类物质会导致细胞DNA损伤,破坏细胞膜甚至打破细胞内氧化还原平衡,最终影响菌株的发酵性能[49]。
2.1.2 低分子量脂肪酸存在于木质纤维素水解液中的小分子脂肪酸主要有甲酸、乙酸和乙酰丙酸等。未解离小分子酸以被动扩散形式进入细胞质内并发生解离从而导致胞内pH降低,这时细胞会通过ATP酶将多余H+泵出细胞,而这个过程需要消耗ATP,但厌氧梭菌本身产能较低,因此额外消耗ATP会抑制细胞生长[50]。刘欢欢[48]发现向发酵液中添加0.5 g/L的甲酸时,甲酸-四氢叶酸连接酶表达量相对于对照组在18 h和24 h均下降,从而使C. acetobutylicum的一碳代谢受到严重抑制。Cho等[51]研究了甲酸和乙酸对C. acetobutylicum和C. berijerinckii的影响,发现当甲酸添加量为0.4 g/L和1.0 g/L时,丁醇和总溶剂产量为对照组的77%和25%,但添加3.7−9.7 g/L的乙酸可促进ABE的发酵,而当乙酸浓度为11.7 g/L时,ABE的发酵性能明显下降。
2.1.3 酚类化合物在木质纤维素水解过程当中,部分木质素会被降解产生琥珀酸、香草醛、对羟基苯甲酸、丁香醛、p-香豆酸和阿魏酸等物质。不同酚醛类物质对细胞的毒害作用机制不同,这取决于其特定的酚类基团。Cho等[52]研究发现酚类化合物在产酸阶段会干扰乙酰辅酶A到丁酰辅酶A的代谢过程,从而影响溶剂产生。Ezeji等[9]研究发现阿魏酸对C. beijerinckii BA101的毒性最大,其次是p-香豆酸,在0.3 g/L阿魏酸或者0.5 g/L p-香豆酸条件下,ABE发酵完全停止。
目前研究发现经酸或蒸汽爆破等方法水解纤维素类物质后均含有上述一类或多类抑制物,虽有报道产溶剂梭菌对低浓度的抑制物具有一定耐受性(如当糠醛和HMF浓度为1.98 g/L时,不会抑制C. beijerinckii A 101的生长[9]),但一般情况下经预处理所获得水解液中抑制物浓度往往处在较高水平,这增加了后续的脱毒处理或ABE发酵的困难。为使抑制物浓度维持在较低水平,减少后续的脱毒工艺,将来需采用更多新的预处理方法,如酶法与其它物理方法相偶联等[53]。
2.2 水解液的脱毒为使工农业废弃物能够成为ABE发酵的原料,需要对水解液进行脱毒处理。目前常用的脱毒方法有化学法脱毒、物理法脱毒、酶法脱毒和原位脱毒等4种方式。
2.2.1 化学法脱毒化学法脱毒是指利用碱性物质或氧化性物质对水解液进行中和或氧化。Mu等[54]比较了Ca(OH)2、NaOH和Na2SO3对经蒸汽爆破预处理的玉米杆水解液进行脱毒,发现以经Ca(OH)2脱毒的水解液进行ABE发酵,总溶剂产率最佳为0.16 g/g,而以经Na2SO3脱毒的水解液进行ABE发酵,总溶剂产率为0.07 g/g。Qureshi等[55]在大麦杆水解液中添加Ca(OH)2可以显著提高ABE发酵过程中总溶剂的产量。利用Ca(OH)2脱毒是因为Ca(OH)2可以中和水解液中的低分子量脂肪酸,且中和形成的不溶性盐可以部分吸附抑制物质,可在离心过程当中被去除,且Ca2+可以增强细胞膜的稳定性。
2.2.2 物理法脱毒常用的物理脱毒方法有阴离子交换树脂、电渗析和活性炭吸附等。阴离子交换树脂可以通过交换离子将呋喃和酚类化合物去除。电渗析是指通过膜将盐类物质去除,活性炭可以吸附水解液中的抑制物质(糠醛、HMF等)。Qureshi等[56]利用电渗析的方法对用碱性过氧化物麦草水解液中的抑制剂(盐)进行处理,结果表明以此处理液为原料能够成功进行ABE发酵。Li等[57]比较了不同脱毒方法对经乙醇有机溶剂预处理松木屑水解液进行脱毒处理,发现利用阴离子交换树脂对水解液进行脱毒后效果最好,经ABE发酵丁醇和总溶剂的产量分别达到6.9 g/L和10.38 g/L。
2.2.3 酶法脱毒常用于木质纤维素水解液脱毒的酶有甲酸脱氢酶和漆酶。漆酶可将水解液中的酚类物质氧化,甲酸脱氢酶可氧化甲酸,以减少甲酸对菌体的伤害。王风芹等[58]利用漆酶和甲酸脱氢酶对玉米秸秆水解液进行脱毒,发现添加5 U/mL的漆酶时,发酵结束后丁醇产量最高,再继续添加漆酶丁醇产量会降低,当添加1 U/mL的甲酸脱氢酶时,可使水解液中甲酸浓度降低61%。
2.2.4 原位脱毒原位脱毒是指菌体自身对水解液中抑制物质进行氧化或还原。Ujor等[59-60]发现在含有糠醛的发酵培养基添加甘油时,可使C. beijerinckii NCIMB 8052的NADH和NADPH水平分别提高1.8倍和3.0倍,在添加别嘌呤醇时虽然不能提高NADH和NADPH水平,但使黄嘌呤和次黄嘌呤磷酸核糖转移酶的信使RNA (mRNA)水平增加2.4倍和6.7倍。甘油促进脱毒是因为甘油与葡萄糖相比在分解代谢过程中会额外产生2 mol的NADH,这使得菌体在产酸过程中迅速将糠醛和HMF还原为毒性较小的物质,从而使发酵不会受到抑制[59]。别嘌呤醇促进脱毒是因为其能够增强菌体对DNA的复制和修复,且促进菌体将糠醛转化为糠醇[60]。
利用不同的脱毒方法对水解液进行处理,均可在一定程度上改善ABE的发酵性能,提高丁醇产量。但以单一脱毒方式进行脱毒会出现糖分流失、抑制物去除效果不理想等问题,为克服上述问题需要多种预处理方法相偶联或建立新的方法对水解液进行脱毒,以减少糖分的流失及增强对抑制物去除效果,最终提高ABE的发酵性能。
3 耐抑制物丁醇生产菌的选育虽然利用各种脱毒方法可使ABE发酵正常进行,但对水解液进行脱毒会增加发酵成本,且会引入新的物质成分,这些物质是否会对发酵造成不良影响还有待更深入地研究,因此,通过驯化、诱变和分子生物学方法等手段筛选获得能够耐抑制物的菌株就显得十分重要。
3.1 通过诱变、驯化等方法获得耐受性高的菌株Guo等[61-62]采用低能离子束注入以及利用连续培养等方法筛选出具有较高耐受性的突变株C. beijerinckii IB4和C. beijerinckii RT66,当以上述突变株为生产菌株,以经稀酸处理过的玉米半纤维素水解液为发酵原料时,其总溶剂产量分别达到9.5 g/L和12.9 g/L,而原始菌株C. beijerinckii NCIMB 8052在此条件下虽然生长良好,但ABE发酵的发酵过程受到抑制。Liu等[21]利用C. beijerinckii 4259为出发菌株,经甲基磺酸乙酯诱变后获得突变株C. beijerinckii ATCC 55025,并以此菌株为生产菌株,利用经烯酸预处理过的麦麸为原料进行ABE发酵,在发酵72 h后丁醇和总溶剂产量分别达到8.8 g/L和11.8 g/L。
蔡的等[63]以C. acetobutylicum ATCC 824为出发菌株,对其进行耐糠醛驯化,最终获得糠醛耐受浓度为3 g/L的菌株,并以此菌株为生产菌株,利用含有3 g/L的糠醛培养基进行ABE发酵,发酵结束后丁醇和总溶剂产量分别为11.09 g/L和20.37 g/L。
3.2 通过分子生物学方法获得耐受性高的菌株Agu等[64]对能利用高浓度甘油的Clostridium pasteurianum DSMZ525中的dhaD1和gldA1 (编码甘油脱氢酶)以及dhaK (编码二羟基丙酮激酶)进行克隆,并在C. beijerinckii中进行高效表达,获得突变菌株C. beijerinckii-gldh,当上述突变菌株在含有6 g/L的糠醛的培养基中进行ABE发酵时,其与原始菌株相比丁醇产率和总溶剂浓度分别提高了46%和40.2%。
4 问题与展望与其他生物燃料(如乙醇和生物柴油)相比,丁醇是一种更为优越的新一代生物燃料。然而传统的丁醇发酵过程往往是以各种淀粉类或葡萄糖类等为底物进行生产,近年来随着全球粮食价格的不断攀升,利用粮食类作物作为丁醇生产原料会增加ABE发酵的成本,同时也存在与人争粮的问题,从而使其无法与石油衍生法相竞争。因此,为提高ABE发酵工业的竞争力,以廉价易得的可再生原料为底物进行丁醇生产是提高其经济竞争力的有效途径之一,而以可再生木质纤维素生物质为原料进行发酵生产丁醇是确保生物丁醇经济竞争力的最佳解决方案。
由于产溶剂梭菌不具备直接利用木质纤维素的能力,因此,以木质纤维素为原料进行ABE发酵,需先对其进行预处理以获得发酵糖,但在所得水解液中往往还含有抑制菌体生长甚至毒害作用的物质,产生毒性的结果会导致ABE发酵结束时出现低产率、低生产强度以及低转化率等现象。为克服上述问题,可从以下几个方面加强研究:(1)建立新的预处理方法,如酶法与其它物理方法相偶联,这样既能够获得可发酵糖,提高水解效率,又使得水解液中抑制类物质浓度维持在一个较低水平甚至不存在抑制类物质。(2)进一步丰富丁醇产菌谱系,采用一些新的高通量筛选方法并结合理性筛选模型,获得更多丁醇产生菌,如能直接利用纤维素且能厌氧发酵的菌种。(3)进一步扩大丁醇产生菌底物利用谱系,研究更多木纤维素进行ABE发酵的可行性,一方面可以变废为宝,同时有利于获得更多可用于丁醇发酵的底物。(4)可将合成生物学及系统代谢工程等现代生物技术手段应用于生产菌种的改造,并结合高效快速的选育技术,从菌种源头进一步提高其耐受抑制物的能力及具备高效的发酵性能。若能以最便宜和最丰富的木质纤维素生物质为原料制备可发酵糖,以及提高菌株耐受不良环境的能力及生产性能,ABE发酵工业将具有更广泛的应用前景。
