随着社会发展和工业化加剧,环境内分泌干扰物污染日益严重,已引起了全球各界的高度重视。环境内分泌干扰物是指由自然产生或人类生产生活释放到周围环境中,可影响人体和动物体内正常激素的功能及内分泌系统的化学物质,也是21世纪最受关注的一类新型环境污染物[1-3]。雌激素作为最重要的内分泌干扰物,具有难降解、环境留存久、累积放大与三致效应强等特点;环境雌激素污染已被认为是一个严重的全球性公害,亟需寻找高效安全的环境雌激素污染治理与环境修复方法[4-7]。
相比于物理与化学治理方法,生物法被认为是去除环境雌激素和进行环境修复的有效方法。其中,微生物降解利用环境分离的微生物菌株或菌群,能以雌激素类物质作为碳源或营养物质,经过复杂的生理代谢将其转化为低雌激素活性物质,并最终将其完全分解为无害的终产物,消除其环境威胁;具有操作简单、成本低廉且无二次污染等优势,被认为是环境雌激素污染去除的主要和有效手段[8-9]。
尽管已有一系列进行环境雌激素降解菌株筛选与鉴定的研究工作,目前微生物转化雌激素的完整代谢通路与降解分子机制等尚未完全解析。为了进一步高效利用和后续工程改造现有的雌激素降解菌株,推进环境雌激素修复进程,需对现有雌激素降解菌株的降解性质、可能的代谢通路与降解机制进行分析与阐述。本文将对雌激素作用强、污染最严重的4种典型雌激素:雌酮(Estrone,E1)、雌二醇(Estradiol,E2)、雌三醇(Estriol,E3) 和17α-炔雌醇(17α-Ethynylestradiol,EE2) 的微生物降解菌株与其相应代谢研究进展进行总结和介绍[10]。
1 雌激素的结构及性质E1、E2、E3、EE2是一类结构类似的脂溶性化合物,其中E1、E2、E3是天然雌激素,EE2是人工合成雌激素。它们含有相同的“6-6-6-5”四环碳骨架结构;E3比E2在C16位多了一个羟基,雌激素活性较低;而EE2则在E2的C17位加入了一个乙炔基,结构更稳定,雌激素效应更强,也更难以被降解[11-12]。四种雌激素的区别仅在于D环上C16、C17位所接官能团的不同(表 1)。

目前筛选获得的环境雌激素降解菌株主要是细菌,大多分离于活性污泥、堆肥、农田土壤或工厂废水中,包括了鞘氨醇单胞菌属(Sphingomonas spp.)、红球菌属(Rhodococcus spp.)、欧洲亚硝化单胞菌(Nitrosomonas europaea)和假单胞菌属(Pseudomonas spp.)等。部分真菌和藻类也被报道具有雌激素降解作用。不同菌株降解雌激素的底物、降解效率、降解周期、转化产物等降解性质有所不同,具有各自的特点(表 2)[8, 13-30]。

天然雌激素中,雌二醇的生理效能最强,环境中分布最广,被认为是最主要的环境雌激素污染物[31]。深入研究微生物降解转化雌二醇机制有助于推进菌株的实际应用与工程改造。
目前对天然雌激素微生物降解机制的研究多集中于细菌对雌二醇的有氧分解过程。早在1966年,Coombre等提出E2的降解首先从A环裂解开始[32];但是这一假设在后续研究中并未被观察到。根据不同微生物代谢雌激素的中间产物,目前推测的细菌代谢雌二醇途径主要有4种,如图 1所示。
(1) 雌二醇首先在C17位被脱氢形成酮基,生成低雌激素活性的E1;之后在C16位开环并在C15、C16位发生羟基化生成内酯,最终将D环裂解[33]。
(2) 因在红球菌和鞘氨醇单胞菌转化E1时观察到4-OH-E2和4-OH-E1的生成,研究者认为微生物降解E2是先从其C4位的羟基化和C14位的脱氢反应开始,生成4-OH-E1后再进一步分解[20]。
(3) 雌二醇的降解可从B环的C7或C8的羟基化开始,在C6或C7位脱氢生成醛基后在C17位脱氢生成酮基,再进行后续降解[30]。
(4) 根据亚硝化单胞菌的研究结果,E0是该菌株转化E2的第一步中间产物,据此推测其雌二醇降解是通过去除其D环C17位羟基并在C16和C17间生成双键来启动的[34]。
此外,研究者在研究鞘氨醇单胞菌代谢EE2时提出E1的另一条代谢途径,即E1的代谢首先从其B环和A环的裂解开始(详见EE2的降解代谢途径1)。根据农场污水中雌激素的有氧和厌氧代谢研究,研究者认为17α-E2-3-硫酸铵盐很可能是其它雌激素物质的前体,经过氧化和前期解离反应生成E1后再进入下一步降解代谢,而且有氧代谢是整个污水处理过程中去除雌激素的主要途径[35]。另外,尽管雌三醇的雌激素活性较低,但它是最难被降解的天然雌激素,目前认为E3分解也是通过其D环的C17位脱氢来启动的[13]。同时研究者也发现,合适浓度的某些可溶有机物可作为雌激素降解菌的最终电子受体,缩短环境中雌二醇的半衰期,实现其氧化与去除[36]。
除了细菌可代谢雌二醇,藻类和真菌对天然雌激素也有一定的降解作用。E1和E2可在藻类中进行相互转化,E1还可生成E3和羟雌甾酮[29]。锰过氧化物酶和漆酶被认为在白腐真菌降解天然雌激素过程中起重要作用[24],而漆酶则可通过将E2氧化为E1或形成E2的二聚体和三聚体,实现E2的代谢转化[22-23]。此外,细胞色素P450也可能参与了真菌对甾醇类激素的代谢过程[25]。
4 17α-炔雌醇的微生物代谢机制EE2是在天然雌激素E2的C17位引入了一个乙炔基,是避孕药的主要有效成分,其化学性质稳定,生理效应强,环境分布广[11-12]。相较于天然雌激素,EE2极难被生物降解,很多天然雌激素降解菌株并不能代谢转化EE2[9]。细菌中目前仅发现鞘氨醇单胞菌、欧洲亚硝化单胞菌和假单胞菌可降解EE2[14, 16-17, 21, 37-38]。根据其代谢产物推测的代谢途径主要有三条(图 2)。
(1) 通过对鞘氨醇单胞菌代谢雌激素物质的产物分析,研究者认为其EE2代谢是从将EE2转化为E1开始,之后在其B环C9和C10位间开环,并在C9位引入酮基;然后A环在C4和C5位间开环,并在C4和C5位上各引入一个酮基;最后以两种可能的方式断裂,再进行下一步代谢[21]。这一推测的EE2代谢过程与已报道的睾丸酮丛毛单胞菌降解类固醇激素的过程相似[37-38]。
(2) 与鞘氨醇单胞菌不同,亚硝化单胞菌代谢EE2的过程被认为是从其A环裂解开始[16]。
(3) 研究者根据欧洲亚硝化单胞菌代谢EE2的产物提出,其EE2的降解首先从C17位乙炔基的降解转化开始,羧基化在这其中起着重要作用[14]。
此外,研究者在对比氨氧化细菌和异养细菌对EE2的代谢产物发现,尽管两者代谢EE2的方式不同,但都生成4-OH-EE2和sulfo-EE2的中间产物,且氨氧化细菌催化这一反应的速度是异养细菌的5倍;但是氨氧化细菌无法继续代谢,其进一步降解是由异养细菌完成的。这暗示着微生物在分解难降解污染物时会采取共同代谢的方式来实现其有效利用[15]。但是,抑制硝化作用进而影响氨氧化细菌活性后,硝化污泥中EE2的代谢速率并没有被显著影响,这说明氨氧化细菌可能不是EE2代谢菌群中的核心菌株[39]。同时,共代谢作用在其他EE2降解细菌中被观察到。不动杆菌和假单胞菌在降解E1、E2、E3时,可实现EE2的共代谢[8];在假单胞菌中还发现了二价锰离子存在时的EE2共代谢作用[19]。
另一方面,在真菌和藻类中也发现了对EE2具有降解作用的菌株,其中真菌的EE2代谢研究较多。根据鉴定的真菌代谢EE2产生的6种化合物结构分析,研究者提出了真菌转化EE2的3种可能方式(图 3A)。漆酶和锰过氧化物酶被认为是真菌代谢EE2的主要酶。由于抑制细胞色素P450可完全阻断真菌对EE2的降解,推测其参与了EE2的转运过程,从而在EE2分解过程中起重要作用[22]。环氧化物水解酶也可能参与了真菌的EE2代谢[26];茎点霉属真菌也被发现具有EE2的降解作用[27]。相比于细菌和真菌,藻类降解EE2的途径更为多样,研究者分别在羊角月牙藻、四尾栅藻和纤维藻中分离了不同的EE2代谢产物,并据此提出了3种可能的代谢方式[28](图 3B)。
5 微生物降解环境雌激素的研究展望随着环境雌激素污染的日益加剧,微生物降解作为其主要且有效的去除手段越来越受到关注,深入相关研究必将提升整体环境治理的效果。据此,本文提出以下几点展望:
(1) 解析雌激素降解菌的基因组成与代谢机制。研究者们已对微生物降解睾丸酮机制的研究较为深入,但对环境雌激素的微生物降解机制仍知之甚少,亟待进一步研究。现有环境雌激素的降解途径主要是根据微生物代谢雌激素的中间物推测获得,其催化相应反应的基因和关键酶并未被解析。由于现阶段大多数雌激素降解菌株的基因组测序尚未完成,其基因组成与遗传进化等性质仍不明确,大大限制了雌激素代谢机制的解析。随着新技术手段不断涌现,应积极运用基因组学、蛋白组学和代谢组学等新技术手段,确定环境雌激素降解菌中的功能基因和关键酶,解析其代谢通路和降解分子机制。本实验室已完成了两株雌激素降解假单胞菌菌株的全基因组测序,并深入分析了其中一株降解菌的差异蛋白组学,相信可推进相关微生物降解雌激素分子机制的研究和菌株改造的进程[17-18, 40]。
(2) 构建安全可控的环境雌激素高效降解工程菌株。目前已分离到的雌激素降解菌株大多具有独特的代谢功能和高效的降解能力,在环境治理方面具有很高的潜在应用价值,但也应对其可能的生物危害与环境威胁予以重视。一旦这些具有潜在危害的微生物在环境中释放应用并长期定殖后,很可能会导致环境的整体生物群落结构与生物多样性发生改变,引发生物危害。因此,在改进雌激素降解菌株的降解效能时,研究者需构建安全可控的工程菌株,降低其潜在的环境威胁。
(3) 寻找合适的雌激素降解菌株的投放方式。合适的投放方式可使菌株进行长时间高效降解,这是菌株实际应用的关键因素。固定化技术是对菌株的实际施用的有益探索,如利用细胞固定化技术固定雌激素降解菌株,可实现污水中雌激素的高效去除[41-42]。可以通过直接将雌激素降解的关键酶进行固定,也可降低整体环境的雌激素水平[43],或者通过雌激素降解酶与其他物质共同作用,也可实现环境中雌激素的降解[44]。因此,找到一种有效的菌株投放方法,对提高雌激素的去除效果和降低环境风险至关重要。
(4) 寻找和筛选更多的雌激素高效降解菌株。研究者分离的雌激素降解菌大多来自于活性污泥,后续研究应尝试在不同环境下进行分离,例如养殖环境、草场及近海等,以获得更多雌激素降解菌资源。不同来源的雌激素降解菌株可能具有不同的代谢途径,这对于全面深入研究和解析微生物代谢环境雌激素的分子机制具有重要意义。
6 结束语微生物降解是目前最为有效也最具前景的环境雌激素去除方式。虽然现在雌激素降解菌分离和其降解性质上的研究较多,但是微生物代谢雌激素的降解通路与分子机制仍不明确,其分子水平上的研究仍显不足。只有全面深入研究解析微生物代谢雌激素的分子机制,确定其降解基因、关键酶与整体网络,才能加快相关的理论研究与实际应用;同时结合适宜的有效投放方式,最终实现环境雌激素的安全高效微生物降解,消除环境威胁,维护人类的健康与生态系统稳定。
