摘要【目的】 探讨菌株Comamonas serinivorans C35降解木质素的能力。 【方法】 测定Comamonas serinivorans C35在木质素培养基中的生长趋势、化学需氧量的去除、木质素降解率、脱色率和相关酶的分泌。分别采用扫描电子显微镜和傅里叶变换红外光谱分析仪检测木质素降解前后的外观结构和化学键的变化。 【结果】 Comamonas serinivorans C35能够在木质素培养基上生长,在培养7 d后,化学需氧量去除率为44.4%,木质素降解率和脱色率分别为43.57%和42.26%。Comamonas serinivorans C35能够分泌木质素过氧化物酶、锰过氧化物酶和漆酶,粗酶液酶活分别能达到648.4、177.8和70.1 U/L。Comamonas serinivorans C35可以使木质素解聚且对其苯环结构、醚键以及C=O键等具有明显的破坏作用。 【结论】 Comamonas serinivorans C35可以降解木质素,在木质素的生物转化中具有潜在的应用价值。

Abstract: [Objective] The aim of this study was to study the degradation of lignin by Comamonas serinivorans C35. [Methods] Comamonas serinivorans C35 growth was measured. Chemical oxygen demand (COD) reduction was investigated. Lignin degradation and decolorization rates were conducted. In addition, secretion of ligninolytic enzymes were investigated by using lignin mineral salt medium. The changes in lignin structural appearance and the chemical bonds of it before and after degradation were assessed by scanning electron microscope (SEM) and fourier transform infrared spectroscopy techniques (FTIR), respectively. [Results] The ability of Comamonas serinivorans C35 in the presence of lignin as a sole carbon source with 44.4% reduction in COD value after 7 days of incubation. The degradation and decolorization removal reached 43.57% and 42.26%, respectively. It was shown that the maximal enzyme activities of lignin peroxidase, manganese peroxidase and laccase of the studied isolate recorded 648.4, 177.8 and 70.1 U/L, respectively throughout the incubation course. SEM revealed the depolymerization of lignin into smaller pieces and FTIR confirmed the benzene ring structure, C-O-C and C=O bonds of lignin were broken down. [Conclusion] The results of this study indicate that Comamonas serinivorans C35 potentiates its ability not only to utilize lignin as a sole carbon source but also its applicable value in lignin bioconversion.

木质纤维素主要是由纤维素、半纤维素和木质素组成。其中木质素与前二者不同的是,这种高分子芳香族化合物是由3种苯丙烷结构单体(对香豆醇、松柏醇和芥子醇)通过碳碳键和碳氧醚键等方式连接形成的一种无定型复杂酚类聚合物[1]。木质素结构复杂,分子量大,而且具有增强细胞壁强度和黏合纤维抵抗微生物腐蚀等作用[2],这也是微生物利用木质纤维素类生物质能源的主要瓶颈之一。自然界中木质素的降解是真菌、细菌及相应微生物群落共同作用的结果[3],其中细菌因为来源广泛、生长快速、易于大规模应用且在木质素降解过程中发挥重要作用(如解聚、开环和支链改性等[4]),因此在木质素污染的环境治理及生物质能源利用方面有着广阔的应用前景。

目前已有不少报道证明细菌能够降解木质素及其模型化合物,而且发现对模型化合物的降解具有多种不同代谢途径。研究较为清楚的是鞘氨醇单胞菌(Sphingomonas paucimobilis) SYK-6,完成了基因组测序并克隆了相关基因,初步构建了木质素代谢网络途径[5]。Crawford等从土壤中分离得到的绿孢链霉菌(Streptomyces viridosporus) T7A和西唐氏链霉菌(Streptomyces setonii) 75Vi2两株放线菌能降解软木、硬木和草类木质素[6-8]。Chandra等从造纸废水中分离出多株具有木质素降解能力的细菌,包括类芽孢杆菌(Paenibacillus)、硫胺素芽孢杆菌(Aneurinibacillus)和沙雷氏菌(Serratia)等[9-11]。Cai等从三国时期的吴国竹简中分离出能降解木质素的细菌有新鞘脂菌(Novosphingobium) B-7、丛毛单胞菌(Comamonas) B-9和贪铜菌(Cupriavidus) B-8等[12-14]。本文研究对象Comamonas serinivorans C35 (GenBank登录号:JN604116) 是从山东省郓城县某农场小麦秸秆堆肥中筛选到的一株丛毛单胞菌[15],它可利用木质素、对香豆酸、阿魏酸、芥子酸、香草醛和对羟基苯甲酸等物质作为唯一碳源生长,本文研究了其对木质素的降解性能。

1 材料与方法 1.1 主要试剂和仪器

碱木质素(alkali lignin)、藜芦醇、2, 6-二甲氧基苯酚和2′-联氨-双-3-乙基苯并噻唑啉-6-磺酸均购自西格玛公司;实验所涉及试剂均为分析纯。AVANTIJ-E型大型高速冷冻离心机、DU-800型紫外/可见分光光度计,美国BECKMAN公司;XY-FD-200F型真空冷冻干燥机,上海欣谕仪器有限公司;XL-30ESEM型环境扫描电子显微镜,荷兰PHILIPS公司;NEXUS 470型傅里叶变换红外光谱仪,美国NICOLET公司。

1.2 培养基的配制

Luria-Bertani培养基配制参考文献[16],pH 7.0-7.2;木质素培养基(g/L):K2HPO4 1.00,MgSO4 0.20,CaCl2 0.10,FeSO4 0.05,MnSO4 0.02,KH2PO4 1.00,(NH4)2SO4 2.00,碱木质素0.50,pH 7.0-7.2,1×105 Pa灭菌20 min。

1.3 方法

将实验室保藏的Comamonas serinivorans C35菌株接种于Luria-Bertani培养基中,30、200 r/min培养至OD600为1.2左右,5 000 r/min离心菌体5 min,用生理盐水洗涤2次后悬浮,取菌液接种于木质素培养基中,连续培养7 d,每隔24 h取培养液,测定菌株的生长状况,而培养液经12 000 r/min离心10 min后的上清液用于COD、木质素降解率、脱色率以及酶活等的测定,未接菌培养基作为空白对照,所有实验做3个平行。

1.3.1 菌株的生长及COD的测定: 通过培养液在600 nm处的吸光度来反应菌株的生长状况,COD的测定采用重铬酸盐法(GB11914-89)。

1.3.2 木质素降解率及脱色率的测定: 280 nm处的吸光度作为木质素浓度测定指标[17]

第n天木质素降解率(%)=(A0-An)/A0×100

A0:接菌前培养基中木质素的浓度;An:接菌培养n天后培养基中木质素的浓度。

465 nm处的吸光度作为色度测定指标[18]

第n天木质素脱色率(%)=(B0-Bn)/B0×100

B0:接菌前培养基的色度;Bn:接菌培养n天后培养基的色度。

1.3.3 酶活的测定: 培养液离心后的上清液作为粗酶液测定Comamonas serinivorans C35分泌木质素过氧化物酶、锰过氧化物酶和漆酶的能力。以藜芦醇为底物测定木质素过氧化物酶酶活,检测波长为310 nm [ε310=9 300 L/(mol·cm)]。测定体系为3 mL,体系中含有50 mmol/L pH为3.0的HAc-NaAc缓冲液、1 mmol/L藜芦醇和100 μL粗酶液,反应在加入1 mmol/L H2O2后启动,30反应5 min,每隔1 min测定310 nm下的吸光值,取吸光值线性变化部分,定义每分钟1 μmol底物转化为产物所需的酶量为一个国际制酶活单位(U)[13]

以2, 6-二甲氧基苯酚为底物测定锰过氧化物酶酶活,检测波长为469 nm [ε469=49 600 L/(mol·cm)]。测定体系为3 mL,体系中含有50 mmol/L pH为3.0的HAc-NaAc缓冲液、1 mmol/L MnSO4、1 mmol/L 2, 6-二甲氧基苯酚和100 μL粗酶液,反应在加入1 mmol/L H2O2后启动,其余同上。

以2′-联氨-双-3-乙基苯并噻唑啉-6-磺酸为底物测定漆酶酶活,检测波长为420 nm [ε420=36 000 L/(mol·cm)]。测定体系为3 mL,体系中含有50 mmol/L pH为3.0的HAc-NaAc缓冲液和1 mmol/L 2′-联氨-双-3-乙基苯并噻唑啉-6-磺酸,加入100 μL粗酶液启动反应,其余同上。

1.3.4 扫描电镜(SEM)观察: 分别取50 mL未接菌的木质素培养基(空白对照)和接菌Comamonas serinivorans C35培养7 d后的木质素培养基,于12 000 r/min离心10 min,上清液冰冻后放入真空冷冻干燥仪中干燥至恒重,干燥后的样品喷金后进行电镜扫描。

1.3.5 傅里叶变换红外光谱(FTIR)检测: 同1.3.4处理样品,精确称取0.001 g冷冻干燥后的样品和0.l g的KBr在玛瑙研钵中磨成细粉末混合均匀。使用压片机在30 MPa的持续压力下将研磨好的细粉末压制成均匀薄片,将薄片放入傅里叶红外光谱仪上进行扫描,扫描的波数范围为4 000-400 cm-1,采用纯KBr粉末压制成的薄片作为背景样品。

2 结果与分析 2.1 细菌的生长及COD的去除

Comamonas serinivorans C35在木质素培养基中的生长趋势和COD降解情况如图 1所示。从图 1中可以看出,培养前3天菌体生长较快,在第5天OD600达到了最大值0.71,随后进入稳定期,OD600值有所下降。相对应的COD降解曲线表明,前3天COD降解迅速,随后缓慢下降。经过7 d的培养,COD浓度从274.7 mg/L下降到152.7 mg/L,去除率达到了44.4%。以上结果表明,Comamonas serinivorans C35可以将木质素作为唯一碳源进行生长,并且能显著降解木质素,且COD的下降主要发生在对数期和稳定期,表明菌株对木质素的降解主要是在初级代谢阶段,这与Ramachandra等[19]的研究结果一致。

2.2 木质素降解率及脱色率

菌株Comamonas serinivorans C35对木质素降解率及脱色率随培养时间的变化如图 2所示,从图 2中可以看出,木质素降解率和脱色率在培养的前5天显著增加,在第5天的时候降解率和脱色率分别达到38.53%和40.48%,随后变缓慢直至稳定,在培养7 d时降解率和脱色率分别达到43.57%和42.26%。结果表明Comamonas serinivorans C35能有效地对木质素进行降解、脱色。

2.3 酶活的测定

图 3为该菌株分泌3种胞外酶的能力,从图 3中可以看出,Comamonas serinivorans C35具有较强的木质素过氧化物酶分泌能力,在前3天酶活增加较快,且在第4天达到最大值648.4 U/L,随后降低。然而分泌锰过氧化物酶和漆酶的能力相对较弱,尤其是漆酶,在培养3 d后达到了最高酶活,仅有70.1 U/L,锰过氧化物酶也在第3天达到最大值177.8 U/L,随后缓慢降低。这3种酶合成是在对数期和稳定期内,且酶活变化趋势与Comamonas serinivorans C35在木质素培养基中的生长规律基本一致。

2.4 扫描电镜(SEM)分析

利用扫描电镜观察木质素降解过程中的形貌变化,是目前证明木质素能否被微生物修饰或降解的重要技术手段之一,木质素样品在Comamonas serinivorans C35降解前后的形态放大相同的倍数(500×)后,可以看出未经处理的木质素样品(图 4A)结构致密且呈块状,而经Comamonas serinivorans C35处理7 d后的样品(图 4B)结构呈疏松多孔状。通过对比图 4AB可证明Comamonas serinivorans C35能够对木质素进行解聚,破坏木质素结构,使其裂解成细小的碎片。

2.5 傅里叶变换红外光谱(FTIR)分析

木质素样品红外光谱图如图 5所示,可以看出木质素降解前后的红外光谱图发生了较大的变化。

根据木质素的化学结构并参考有关文献[18]可知,3 430-3 370 cm-1处的-OH伸缩振动峰增强且发生了位移,2 970-2 920 cm-1处的-CH3、-CH2伸缩振动峰减弱,这说明木质素苯丙烷结构单体间的C-C键和C-O-C键可能发生了氧化断裂,导致C-H振动减弱,且对侧链基团-OH进行了修饰。经Comamonas serinivorans C35降解后的木质素样品在2 400 cm-1和2 100 cm-1出现峰,这是由于生成C≡C或C=C=C结构产生的伸缩振动。1 590 cm-1处的苯环骨架振动和C=O伸缩振动减弱并发生位移,说明形成了新的苯环醛酮羧酸类物质。侧链的断裂导致了1 420 cm-1处的苯环振动和C-H键面内振动峰增强。1 130 cm-1处的C-O伸缩振动峰变宽,这可能是木质素中醇类、酚类或脂链醚结构发生了变化。1 050 cm-1和982 cm-1处的C-O-C反对称和对称伸缩振动减弱,再次证明部分该键发生了断裂。以上结果表明,Comamonas serinivorans C35可以攻击木质素的苯环结构及其侧链,包括羰基、烯基和苯等不饱和键发色基团以及碳氧醚键等助色基团,同时也可推断Comamonas serinivorans C35能使木质素色度降低的原因是对这些发色基团和助色基团产生了破坏。

3 结论与讨论

木质素是世界上第二丰富的可再生有机资源,但由于其结构复杂,一直未被充分利用。对木质素进行综合利用的方法已进行了几十年的研究,但仍存在许多问题,如化学法会造成二次污染,物理法有高能耗的挑战,而生物法虽对环境友好,但存在效率低的问题。目前木质素大多燃烧或者废弃,因此筛选高效降解木质素的微生物具有现实意义,这不仅充分利用了废弃物,而且对开发以木质纤维素为原料的可再生能源具有重要意义。

本文的研究对象Comamonas serinivorans C35是一株从小麦秸秆堆肥中筛选到的细菌,已有研究表明该属细菌能降解多种环境污染物,如Comamonas testosteroni ZD4-1能降解苯酚、2, 4-二氯苯酚和对硝基苯酚等[20]Comamonas testosteroni KF-1能降解3, 4-磺苯基丁酸盐,生成4-磺基乙酰苯和4-邻磺基苯酚这两种中间代谢产物[21]Comamonas sp. UVS能使直接红等纺织染料脱色分解成对环境无危害的代谢物,且在脱色过程中该菌能分泌木质素过氧化物酶和漆酶[22]Comamonas sp. strain CNB-1能降解硝基苯和对氯硝基苯;Comamonas sp. strain E6可通过原儿茶酸4, 5开环途径降解对苯二酸酯和间苯二酸酯[23-24]Comamonas sp. VS-MH2能降解4种偶氮染料混合物[25]。该属细菌也有木质素降解功能,其中Comamonas sp. B-9能在强碱性条件下降解木质素,可用于造纸废水的处理[26]。本文有效证明了Comamonas serinivorans C35能将木质素作为唯一碳源进行生长。Chandra等报道的3株细菌Aneurinibacillus aneurinilyticusPaenibacillus sp.和Bacillus sp.均不能将木质素作为唯一碳源进行生长,只有在加入适量碳源和氮源后方可利用木质素,其中Aneurinibacillus aneurinilyticus COD去除率、降解率和脱色率分别达到43%、33%和58%,Paenibacillus sp.和Bacillus sp.降解率分别达到37%和30%[9, 27];Cai等报道的3株细菌Comamonas sp. B-9、Novosphingobium sp. B-7和Cupriavidus basilensis B-8能将木质素作为唯一碳源生长,其中Comamonas sp. B-9 COD去除率、降解率和脱色率分别达到32%、45%和54%,Novosphingobium sp. B-7和Cupriavidus basilensis B-8 COD去除率分别达到了34.7%和55.6%[12-14]Comamonas serinivorans C35对COD去除率、降解率与已报道的木质素降解菌相比均达到较高值,表明其具有较强的木质素降解能力,具有进一步研究的意义和潜在的工业价值。细菌分泌木质素过氧化物酶的报道较少,Cai等研究的菌株Comamonas sp. B-9具有较高的锰过氧化物酶和漆酶酶活,分别可达2 903 U/L和1 250 U/L,但也没有检测到木质素过氧化物酶酶活[13],而Comamonas serinivorans C35具有较强的木质素过氧化物酶分泌能力是其区别于其他丛毛单胞菌和木质素降解菌的特点。

本研究表明Comamonas serinivorans C35是一株优良的木质素降解菌,在将来生物质能源利用以及木质素污染的环境治理方面有着潜在的应用价值。下一步对Comamonas serinivorans C35的研究将通过全基因组测序以及转录组或者蛋白组的组学技术,阐明其对木质素的降解机制以及代谢途径,并期望发现具有实际应用价值的木质素代谢相关酶。
