疫苗接种是预防传染病的一种重要策略。然而,目前许多疫苗在生产过程中存在抗原产量偏低的问题,由此导致疫苗的生产成本较高、有效抗原含量低和免疫效果差等问题。为此,研究人员尝试了不同策略来提高病毒疫苗抗原的产量,以改进疫苗的质量并降低生产成本。文中总结了近年来提高疫苗中病毒抗原产量的主要方法,包括改造病毒基因、改善病毒对细胞的适应性、优化抗原表达体系、改进疫苗生产工艺等方面。并分析了不同策略的优点和存在的问题,提出了提高疫苗抗原产量的一些设想。
Vaccination is an important strategy to prevent infectious diseases. However, low antigen yield of vaccine producing strains may lead to high cost of vaccines, low antigen production and vaccine failure. In recent years, many efforts have been made to improve the antigen yield of many vaccines. This mini-review summarizes various methods for increasing the antigen yield for vaccine production, including genetic modification of viruses, improvement of the adaptation of viruses to cells, and optimization of antigen expression systems and manufacturing procedures. Furthermore, we discuss the advantages and the problems of current strategies, as well as indicate the perspectives.

基于丙型肝炎病毒(HCV) JFH1株的感染性细胞培养物(HCVcc)系统对于HCV的研究至关重要。HCVcc系统虽然促进了HCV的研究,但是它不能产生形态学和疫苗研究所需的大量病毒粒子。Mathiesen等将HCVcc在Huh7衍生的肝癌细胞中连续传代产生了具有适应性突变的病毒准种,该适应性突变增强了病毒的组装、特异性感染及其在细胞间传播的能力,从而提高了病毒粒子的产量[
尽管研究证实,病毒可以通过连续传代的方式获得使其复制效率增强的突变,但是该策略具有不确定性和随机性。
所谓易错PCR是指通过调整PCR反应条件增加突变频率,使得错误碱基以一定的频率随机掺入到扩增基因中,从而得到随机突变的DNA群体,最后用合适的载体克隆突变基因[
以上研究表明,血凝素与受体分子的紧密结合可能是病毒高效复制的原因。根据该机制,可以利用易错PCR对其他病毒的受体结合蛋白基因进行突变,以获得可以更好结合细胞的突变体病毒,从而提高疫苗毒株的产量。例如,利用易错PCR对猪瘟病毒Erns基因进行突变,可能会获得与硫酸乙酰肝素(HS)紧密结合的突变体病毒,从而提高猪瘟疫苗的滴度。
流感病毒疫苗株在鸡胚中生长缓慢,造成疫苗生产费时费力[
该策略可以快速获得针对流行毒株的流感疫苗,即将流行毒株的HA和NA基因重组到高产毒株骨架上。但可能存在的问题是,由于流行毒株的HA和NA基因与高产毒株的骨架不匹配而无法获得重组病毒。

对于利用细胞生产的疫苗,高密度细胞培养无疑也是提高疫苗产量的有效策略之一。目前可以用人胚肺成纤维细胞、鸡胚细胞和Vero细胞生产安全有效的狂犬病疫苗。而Vero细胞被认为是生产狂犬病疫苗的最适基质[
经过优化细胞培养反应器、高密度细胞培养,可以有效提高疫苗产量。然而对于某些抗原含量要求较高的疫苗而言,如果没有高效的纯化和浓缩工艺,那么疫苗的效力仍难以得到保障。
Arifin等通过优化细胞培养和病毒离心分离过程提高了新城疫疫苗的产量。首先,作者测试了Vero细胞在不同培养基中的生长情况,结果发现,在DMEM (Dulbecco’s modified Eagle’s medium)培养基中细胞密度最大可以达到1.93×106个/mL;然后用微载体在搅拌式生物反应器中生产新城疫病毒;最后作者通过优化病毒样品的浓度、离心的温度和时间,获得了高滴度的病毒[
总之,改进疫苗生产工艺尤其是细胞培养的策略,或者是优化整个生产过程,可以有效提高疫苗产量。
接种疫苗有助于人类和动物更好地抵御病原微生物的侵害。在疫苗出现之初,许多疫苗是通过将毒株在异源动物体内传代适应所得,如猪瘟兔化弱毒疫苗。但是用动物体生产疫苗存在传播病原的潜在风险。目前,用细胞生产疫苗更加安全、稳定、可控。不过,改造病毒基因和提高病毒对细胞的适应性这两种策略,大部分还处于实验室研究阶段,没有成为商业化疫苗。其原因是一些策略实现难度较大:例如许多病毒的特异性受体还没有得到鉴定;敲除细胞内干扰素通路相关分子策略尚未应用于生产实践,主要由于目前对病毒与宿主细胞内干扰素通路相互作用的研究还处在发展阶段,对其作用机制了解甚少。虽然许多策略在实验室研究中行之有效,但是仍然存在如下问题:1) 具有不确定性。通过改变病毒骨架研发高产流感疫苗株时,如果HA和NA与骨架病毒的6个基因片段不兼容,就很难产生高产种子毒株,而且该方法不能通过选择基因片段来产生高产毒株[
对病毒的吸附、入侵、复制、装配、释放等整个复制周期以及病毒与宿主之间相互作用进行更加深入的研究,有利于开发新的提高病毒疫苗抗原产量的策略。了解流感病毒HA和NA基因为什么能够和高产毒株骨架相适应,也许可以帮助解决某些流感病毒的HA和NA不能和高产毒株的骨架相适应的问题,从而可利用高产毒株骨架制备各种不同的高产毒株。
许多病毒基因组的非编码区与病毒的复制密切相关,因此如果对其进行改造,有望获得高滴度的疫苗株;虽然许多病毒的特异性受体尚未被确定,但可构建过表达已知通用受体的细胞系,例如在广泛用于疫苗生产的Vero细胞上过表达HS等通用受体,可以提高病毒抗原产量;腺病毒甲病毒复制子嵌合载体或许可以应用于除猪瘟病毒外其他病毒保护性抗原基因的高效递送和表达;而在利用细胞生产疫苗时,细胞培养环节可以相互借鉴,我们可以开发出一套最优化的细胞培养系统用于多种疫苗的生产。此外,除了提高疫苗抗原的产量,还可以提高其质量(特别是免疫原性)。
